Summary:

The Translational Core Laboratory consists of the Specimen Collection, Processing and Point of Care, Biochemistry, Cell Culture/DNA Isolation, and Molecular Biology core laboratories. Laboratory testing is integrated across Penn and CHOP, and TCL services are provided at multiple physical locations at both Penn and CHOP.

Instruments

  • ABI PRISM 7900HT sequence detection system ( Real-time PCR machine )

    "The 7900HT System is a high-throughput real-time PCR system that detects and quantitates nucleic acid sequences. An Automation Accessory combined with 384-well plate capability make the 7900HT system ideally suited to meet the high-throughput requirements of today's drug discovery process. Key applications include gene expression quantitation and the detection of single nucleotide polymorphisms (SNPs) using the fluorogenic 5' nuclease assay."

  • COBAS 6000 c501 biochemical analyzer ( Chemistry analyzer )

    This is an automated analyzer capable of executing high throughput analysis on a variety of samples from humans and non humans. Sample types accepted include serum, plasma, urine, CSF and others. More than 80 different biomarkers can be analyzed using a wide spectrum of biochemical assays. Assays are all validated as FDA in vitro diagnostic category.

  • COBAS e411 automated immunochemical analyzer ( Chemistry analyzer )

    The COBAS e411 is capable of full automated operation and offers a menu of nearly 30 assays that are all FDA certified for use as in-vitro diagnostic procedures. This instrument automates the 2-site 'sandwich' immunoassay by using combination of magnetic beads and electrochemiluminescent detection to quantify a variety of ligands including hormones, tumor markers, and other proteins and antigens.

    This stand alone instrument completely automates the traditional 2-site immunoassay and incorporates a fluorescent tag whose signal is digitized and correlated with concentration of immunoassay target. This combination yields detection limits that are nearly 2 two orders of magnitude lower than traditional sandwich ELISAs. The SIMOA offers an established menu of analytes that includes cytokines and growth factors. In addition, user-defined methods may be created and performed on the SIMOA.

  • Sysmex XT-2000iV Automated Hematology Analyzer ( Hematology analyzer )

  • SOMAscan ( Protocol )

    SOMAscan is an aptamer-based high throughput screening protocol that simultaneously screens for 1129 biomarker candidates. From every sample in a group of 24 unknown samples, the SOMAscan process consumes just 65 uL of serum or EDTA plasma. The SOMAscan process is completed within 48 hours from start and yields an output file that contains concentration-related signals for all 1129 targets as quantified within each sample in the group of 24. Completed output files can then be submitted for further bioinformatic data analysis.

    "GeneMapper® Software is a flexible genotyping software package that provides DNA sizing and quality allele calls for all Applied Biosystems electrophoresis-based genotyping systems. This software specializes in multiapplication functionality, including amplified fragment length polymorphism (AFLP®), loss of heterozygosity (LOH), microsatellite, and SNP genotyping analysis. GeneMapper® Software can help users increase data processing efficiency with remote auto-analysis and command line operation, and allows for multiuser, client-server deployment. The software uses Process Quality Values (PQVs) for automated identification that reduces data review time for high throughput genotyping. In addition, the security and audit features help users meet 21 CFR 11 requirements."

    "MacVector is a comprehensive Macintosh application that provides sequence editing, primer design, internet database searching, protein analysis, sequence confirmation, multiple sequence alignment, phylogenetic reconstruction, coding region analysis, and a variety of other functions. MacVector is widely regarded as the most intuitive, easy to use program available for sequence analysis.
